background: it has been suggested that organophosphates may inhibit gamma-aminobutyric acid (gaba) metabolism in synaptosomal preparations. in the present investigation, we have assessed the interaction between paraoxon and the gaba system at synaptic level.   methods: synaptosomes were prepared from male wistar rats (200-250 g). cerebral cortex was dissected and homogenized, then centrifuged at 1000g for 5 min and again at 12000g for 20 min. the pellet containing synaptosomes was resuspended in the buffer solution and the protein concentration adjusted at 1 mg/ml. determination of lactate dehydrogenase (ldh) activity, as a biochemical index for synaptosomal integrity, was assessed. cholinesterase activity of synaptosomes was also determined. synaptosomes were preincubated for 20 min with two concentrations of paraoxon (10 -9 -10 -3 m), and then incubated with [ 3 h]gaba for 10 min, before being washed through 0.65 μm filters.   results: paraoxon inhibited cholinesterase activity of synaptosomes in a concentration dependent manner. synaptosomal accumulation of [ 3 h] gaba/gaba was time dependent and peaked at 15 min. paraoxon significantly increased the uptake in nano molar concentrations and decreased it at higher concentrations.   conclusion: the result of this study indicate that in synaptosomes prepared from rat cerebral cortex paraoxon increases gaba uptake at low dose and inhibits its uptake at high doses. this may imply a role for organophosphate-induced convulsion, which needs further clarification. iran j med sci 2006; 31(3):125-130.   keywords ● synaptosome ● gaba ● organophosphate ● paraoxon

Effect of paraoxon on the synaptosomal GABA uptake in rat hippocampus and cerebral cortex

Introduction: Paraoxon (the neurotoxic metabolite of organophosphorus (OP) insecticide, parathion) exerts acute toxicity by inhibition of acetylcholinesterase (AChE), leading to the accumulation of acetylcholine in cholinergic synapses and hence overstimulation of the cholinergic system. Since, reports on changes in the level of γ- amino butyric acid (GABA) during OP-induced convulsion have bee...

Effect of paraoxon on the synaptosomal GABA uptake in rat hippocampus and cerebral cortex

Introduction: Paraoxon is the neurotoxic metabolite of organophosphorus (OP) insecticide parathion and causes acute toxicity by inhibition of acetylcholinesterase (AChE). AChE inhibition leads to the accumulation of acetylcholine in cholinergic synapses and hence results in an overstimulation of the cholinergic system. Reports on changes in the level of γamino butyric acid (GABA) during OP-indu...

Non-cholinergic effects of paraoxon on [3h]-GABA release from rat cerebellar giant synaptosomes

Diethyl p-nitrophenyl phosphate (paraoxon) is the active toxic metabolite of parathion. Some evidences indicate that OPs affect the GABA system via noncholinergic mechanisms. The purpose of this study was to investigate the effects of paraoxon on K+-evoked [3H]-GABA release from cerebellar synaptosomes. Adult male rats (200 ± 30 g; 3-4 months old) were sacrificed by decapitation and the cerebel...

The inhibitory effect of Paraoxon on cerebellar synaptosome GABA uptake in rats

Introduction: Compounds which are used to treat organophosphate (OP) poisoning are not able to fully alleviate long lasting effects. They are mainly used to antagonize cholinergic effects of Ops. However, non-cholinergic effects, such as interference with different neurotransmitter systems, especially GABA release and uptake, are recently attracting more attentions. We have tried to investi...
